Project Overview

To investigate the effects of thickness, UV radiation exposure, and RGD adhesion molecules on the viability and testosterone production of human prostate cancer cells embedded within polyethylene glycol diacrylate hydrogel. The overall goal of this project is to design an encapsulation system that offers efficient immunoprotection and effective diffusion of oxygen, nutrients, hormones, and metabolic wastes. Conceptually, the stated encapsulation system, along with embedded human prostate cancer cells, will enable the restoration of un-regulated testosterone levels commonly observed in elders, and retard the symptoms of aging.

Project Timeline

Week Reporting Period Beginning Activities
1 January 26 Selected team roles and e-mailed information to advisor. Materials search for UV lamp. Brainstormed outreach activities and content of presentation.
2 February 2 Materials search for dialysis tubing. Diacrylation of MW 12000 PEG. Worked on outreach presentation.
3 February 9 Performed dialysis and lyophilization of PEGdA samples. Worked on outreach PowerPoint presentation.
4 February 16 Set up appointment for outreach activity. Tested the intensity and effectiveness of Miguels UV lamp.
5 February 23 Prepared for outreach presentation. Contacted Spectronics Corporation about interchangeability of their UV tubes.
6 March 2 Client ordered replacement 365nm UV tubes from Spectronics Corporation. Performed swelling ratio experiments. Took a new team photo. Carried out outreach activity on Wed, 3/7 at Madison East HS. Preparing for mid-semester presentation.
7 March 9 Design notebooks due Wed. 3/14/07 by 4pm.
8 March 16 Prepared additional PEGdA samples. Planned for subsequent hydrogel swelling experiment. Scheduled meeting on Mon. 3/26 with Dr. Tim Stiles to measure pre-swelled hydrogel thickness via ultrasound.
9 March 23 Client meeting with Dr. Atwood on Friday 3/23 at 3pm. UV exposure experiments for swelling ratio from 13 to 15 min. Began paperwork for gaining access to Dr. Atwoods lab..
10 March 30 Spring Break
11 April 6 Acquired identification badges and card activation for access to lab. Received brief training on cell culture specifics from Miguel and Andrea from Dr. Atwoods lab. Hydrogel thickness measurements via ultrasound by Dr. Tim Stiles.
12 April 13 Continued to work on outreach report sections. Confirmed pre and post-swell hydrogel thicknesses at 220 and 340 um, respectively.
13 April 20 Tested effectiveness of new 365 nm tubes obtained from Spectronics Corporation for our application. Began hydrogel thickness experiements (8 thicknesses w/3 replicates each).
14 April 27 Complete experimentation: Progesterone release (Progesterone ELISA), hydrogel thickness verification (ultrasound), and cell viability of encapsulated PC3 cells (Cell Titer-Blue Assay).
15 May 4 Complete final presentation, final paper, and design notebooks for evaluation.
